一般的なライゲーション反応に適した範囲で、目的のインサート/ベクターモル比にするために必要なインサート量を計算します。

インサートDNA所要量 [g] = ベクターDNA量 [g] x {インサートDNA長/ベクターDNA長} x 目的のインサート/ベクターモル比

注：モル比は{1, 2, 3, 5 ,7}から選択します。

インサート長はベクター長よりも小さい値にします。

最初に、インサートDNAとベクターDNAを制限エンドヌクレアーゼで消化し、粘着末端（または平滑末端）を持つDNA断片を生成します。次に、目的のDNA断片を切り取った後に結合し、DNAリガーゼで処理します。最後に、得られたインタクトなベクターをコンピテントなE. coli細胞に形質転換します。形質転換した細胞は適切な遺伝子選別方法によって識別します。
