The RNeasy DSP Midi Kit uses silica-membrane technology (RNeasy technology) for manual isolation and purification of total RNA from human blood and bone marrow aspirates for use with in-vitro diagnostic QIAGEN products that list the RNeasy Midi Kit as an accessory.
The RNeasy DSP Midi Kit uses well-established RNeasy technology for purifying RNA. RNeasy technology simplifies total RNA isolation by combining the stringency of a guanidine-isothiocyanate lysis with the speed and purity of silica-membrane purification.
With the RNeasy DSP Midi Kit, total RNA is easily purified from human blood or bone marrow aspirates. Erythrocytes from blood are lysed (erythrocyte lysis buffer not included in the kit, Buffer EL, cat. no. 79218) and white blood cells are collected, lysed, and homogenized. Ethanol is added to provide ideal binding conditions and the lysate is then loaded onto the RNeasy Midi column. Subsequent washing and centrifugation steps are applied before RNA is eluted in RNase-free water. Detailed protocols can be found in the instructions for use of the corresponding downstream applications.
RNA purified using the RNeasy DSP Midi kit can either be directly used for sensitive analytical detection in downstream assays such as qPCR or RNA can further be concentrated using the RNeasy DSP MinElute Cleanup Kit.
