QIAsymphony DSP Virus/Pathogen Kits, in combination with the QIAsymphony SP, enable automated purification of viral nucleic acids and bacterial DNA from a broad range of sample materials for in vitro diagnostic use. Kits are available in mini and midi format for sample volumes of 200 μl and up to 1000 μl. Optimized protocols provide high yields of pure nucleic acids, and flexible elution volumes allow nucleic acid concentration to be easily optimized for each downstream application. Novel, prefilled reagent cartridges reduce setup time and minimize manual handling. Bar code reading enables full tracking of reagents.

Principle

QIAsymphony DSP Virus/Pathogen Kits can be used to purify viral nucleic acids from serum, plasma, or CSF, and viral nucleic acids and bacterial DNA can be purified from respiratory samples such as swabs, as well as urine and urogenital swabs (cervical and urethral). The kits can be used to purify nucleic acids from a broad range of DNA and RNA viruses, as well as bacterial DNA from Gram-negative and Gram-positive bacteria.

QIAsymphony DSP Virus/Pathogen Kits combine the speed and efficiency of silica-based purification of viral and bacterial nucleic acids with the convenient handling of magnetic particles (see " Schematic of the QIAsymphony SP principle"). Innovative, ready-to-run reagent cartridges are prefilled with all reagents required for the purification procedure and are automatically opened by the QIAsymphony SP, which helps to minimize the risk of environmental contamination (see figure " Prefilled, sealed reagent cartridges"). Optimized protocols provide high quality nucleic acids with efficient removal of proteins, nucleases, and other impurities. Nucleic acids purified with QIAsymphony DSP Virus/Pathogen Kits are ready for direct use in downstream applications (e.g., PCR). The user can choose between different elution volumes.

Procedure

The purification procedure is designed to ensure safe and reproducible handling of potentially infectious samples, and comprises 4 steps: lyse, bind, wash, and elute (see " QIAsymphony DSP Virus/Pathogen procedure"). Worktable setup is rapid, which saves your valuable time. Simply place up to 2 reagent cartridges in the consumables drawer. The reagent cartridges can be either from the same kit or from different kits, enabling different purification procedures to be performed within the same run of 96 samples. Reagent volumes only for the selected number of samples are used, giving you complete cost control.

Applications

QIAsymphony DSP Virus/Pathogen Kits enable sensitive detection of a broad range of DNA and RNA viruses, as well as bacterial DNA. Yields are linear, allowing accurate quantitative analysis for both low and high viral or bacterial titers. The high-quality nucleic acids are ready to use in all downstream applications, including sensitive detection assays using quantitative, real-time PCR or RT-PCR.

What sample input formats can be used for the QIAsymphony DSP Virus/Pathogen protocols?
Tube carriers for up to 24 tubes (primary or secondary tubes or tubes containing internal controls) can be used. A list of compatible labware formats for sample input can be found at www.qiagen.com/goto/dsphandbooks. Slots 1–4 are used for samples to be processed. The internal control-carrier RNA-Buffer AVE mixture is loaded into Slot 5.

Why do I have to use an ACS (Assay Control Set)?
The ACS specifies the combination of a protocol and a predefined carrier RNA–internal control mixture. The user can define an internal control mixture using the QIAsymphony Management Console. A predefined internal control-carrier RNA-Buffer AVE mixture can be automatically identified by the instrument using a bar code label or it can be chosen manually.

How much starting material can be used with the QIAsymphony DSP DNA Mini Kit on the QIAsymphony SP?

Tissue: If no information about the expected yield is available, we recommend starting with 25 mg sample material. Depending on the yield obtained, the sample size can be increased in subsequent preparations to a maximum of 50 mg.

FFPE: Up to 4 sections, each with a thickness of up to 10 µm, or 8 sections with a thickness of up to 5 µm and a surface area of up to 250 mm2, can be combined in one preparation.

All complex protocols require Buffer ATL, which is not included in the QIAsymphony DSP Virus/Pathogen Kits. Buffer ATL can be ordered separately (Buffer ATL (4 x 50 ml), cat. no. 939016).

Can the QIAsymphony DSP Virus/Pathogen system be used for diagnostic or prognostic procedures?
Yes. The QIAsymphony DSP Virus/Pathogen Kits are designated “For In Vitro Diagnostic Use”. It is the user’s responsibility to validate system performance for any procedures used in their laboratory that are not covered by the QIAGEN performance evaluation studies. The system performance has been established in performance evaluation studies purifying viral DNA and RNA from human serum, plasma, or cerebrospinal fluid (CSF), and purifying viral DNA and RNA, as well as bacterial DNA, from respiratory and urogenital samples. To minimize the risk of a negative impact on the diagnostic results, adequate controls for downstream applications should be used. For further validation, the guidelines of the International Conference on Harmonisation of Technical Requirements (ICH) in ICH Q2 R(1) Validation of Analytical Procedures: Text and Methodology are recommended. Any diagnostic results that are generated must be interpreted in conjunction with other clinical or laboratory findings.

Which elution volumes can be used for the QIAsymphony DSP Virus/Pathogen protocols?
For all cellfree and complex protocols, an eluate volume of 60 µl, 85 µl, or 110 µl can be selected. These are the minimum accessible volumes of eluate in the final elution tube. The initial elution volumes, which have to be used for the internal control, as well as titer calculations, are 90 µl, 115 µl, and 140 µl.

Do samples require pretreatment before they are loaded onto the QIAsymphony instrument?
Pretreatment can improve the nucleic acid extraction for some sample types. Viscous samples (e.g., sputum, respiratory aspirates) might require liquefaction to enable pipetting. Performance may be improved for Gram positive bacteria by pretreatment with lysozyme. Pretreatment protocols are provided in the QIAsymphony DSP Virus/Pathogen Handbook.
