Procedure

The QIAsymphony RGQ workflow starts with purification of nucleic acids on the QIAsymphony SP instrument. The eluates, containing purified nucleic acids from the sample preparation procedure, are transferred to the QIAsymphony AS module, which performs assay setup. The assays are then manually transferred to the Rotor-Gene Q for PCR and data analysis.

To suit individual workflow requirements, there are 2 modes of system operation: integrated or independent. In the integrated mode, samples processed on the QIAsymphony SP are transferred automatically to the QIAsymphony AS, reducing manual handling steps and documentation. For added flexibility, the QIAsymphony SP and AS can also be operated independently of each other in independent mode. See " QIAsymphony workflows".

FAQ

The QIAsymphony RNA kit uses RLT plus as lysis buffer. Can samples lysed in RLT and then stored in the freezer be used on the QIAsymphony?

The RLT buffer is more viscous than the RLT plus buffer. Extraction will work fine, but there may be some magnetic bead carryover to the eluate.

There is no barcode on the QIAsymphony cooling adapter, and the instrument asks for one. What do you suggest I do?

The barcode on the adapter at the cooling elution position is read by default. This can be changed in the configuration settings which are available from the tools menu if you have logged in as Supervisor. The option is on the tab Process SP1 and is called ”On which elution slots should the bar codes be scanned”. To disable the barcode reading, put the setting to ”none”.

With the QS-RGQ the user can process 1 to 4 batches with 1 to 24 reactions per specific batch. RNA assays are limited to 72 reactions in total.

For PCR:  216 reactions for DNA and 72 reactions for RNA viruses can be performed.
