How can a positive threshold be set when running the QIAxcel System?
For setting a positive threshold when running the QIAxcel System we would recommend using a percent value. A value of 7-10% should eliminate unwanted baseline noise. Just choose the smallest peak height value (RFU value with the pUC18 sample by pointing the curser on top of the peak) and adjust the Positive Threshold (%) to be smaller than the ratio of the two peaks (smallest peak/highest peak).
Note: When performing a calibration, the software normalizes all intensities to 5 RFU (100%).
