The QIAcuity One-Step Viral RT-PCR Kit enables sensitive quantification of RNA targets on the QIAcuity digital PCR instrument.

The kit contains a 4x concentrated PCR Master Mix optimized for microfluidic use in the QIAcuity Nanoplates. The kit enhances the specificity and efficiency of probe-based digital PCR to provide accurate, singleplex or up to 5-plex analysis.

The QIAcuity One-Step RT-PCR Master Mixes for hydrolysis probe-based dPCR use QIAGEN’s latest version of high-quality DNA polymerase. The unique combination of QIAGEN's proprietary and well-proven buffer technology optimized for the nanoplate microfluidics delivers highly consistent results in terms of sensitivity, reproducibility and efficiency.

Detection from single to 5-plex with the QIAcuity Probe PCR Kit
The special master mix in the QIAcuity Probe PCR Kit enables accurate quantification of up to 5 targets having widely differing abundance in a well of the QIAcuity Nanoplate. This saves time and money and reduces the amount of sample material needed. Moreover, the duplex or multiplex PCR data obtained is comparable with that obtained from a singleplex PCR.

The QIAcuity One-Step Viral RT-PCR Kit contains a ready-to-use 4x PCR Master Mix, eliminating the need to optimize reaction and cycling conditions.

Procedure

The QIAcuity One-Step Viral RT-PCR Kit contains a ready-to-use 4x PCR master mix, eliminating the need to optimize reaction and cycling conditions. Simply add the 100x RT mix, template RNA and primer-probe sets to the master mix and follow the procedure in the Quick-Start Protocol to get fast and reliable results on the QIAcuity.

Applications

The QIAcuity One-Step Viral RT-PCR Kit, combined with the QIAcuity Digital PCR System and the QIAcuity Nanoplates, enables quantitative analysis of RNA targets for various applications, including but not limited to viral RNA detection.

Here we report the use of saliva samples in combination with dPCR as a suitable alternative to screen for individuals infected with SARS-CoV-2.
The QIAcuity digital PCR system combined with the QIAcuity One-Step Viral RT-PCR Kit enables precise detection and quantitation of vector-borne viruses in mosquitoes. The results presented in this comparison study showed that digital PCR is a powerful tool for absolute quantitation of low abundant targets and is a more reliable detection method than qPCR. Multiplexing allows detection and quantitation of multiple targets in a single reaction more efficiently by increasing sample throughput at a reduced cost per target.
