QIAamp DNA Investigator Kit可从罪案现场样本等多种法医检材和人类身份鉴定样本中纯化基因组DNA,如干血斑、骨头、性侵犯样本、拭子、滤片等。试剂盒使用QIAamp MinElute离心柱纯化高品质DNA,具有灵活的洗脱体积。纯化过程快速高效,得到的DNA在多种下游应用,如定量PCR和STR分析中表现出色,信噪比高。另外,在QIAcube全自动核酸纯化仪上QIAamp DNA Investigator Kit可自动纯化DNA。

QIAamp DNA Investigator Kit适用于从痕量样本中(如:罪案现场样本)纯化DNA;进行基因分型,包括DNA指纹图谱分析和亲子鉴定;以及对参考样本进行常规分析。

QIAamp DNA Investigator流程高度适用于对多个样本的同时处理。样本裂解后,通过此操作流程在30分钟内即可获得纯化DNA。DNA在ATE缓冲液或纯水中洗脱得到,可即时用于下游反应,或储存于–20°C环境中。纯化得到的DNA不含蛋白质、核酸酶和抑制剂。

原理

QIAamp DNA Investigator Kit具备硅胶膜选择性结合DNA的特性,以及20 µl到100 µl的灵活洗脱体积。

程序

S_0200_IAS_0285QIAamp MinElute离心柱可纯化高品质DNA,洗脱体积灵活。QIAamp DNA Investigator流程包括4步(参见" Procedure")。样本在变性条件下被蛋白酶K裂解。DNA与硅胶膜结合,杂质流出后洗去残留的污染物。最后从硅胶膜上洗脱得到浓缩的纯化DNA。QIAamp DNA Investigator Kit可在QIAcube全自动核酸纯化仪上自动纯化DNA。 

该操作流程经专门设计,可确保样本间无交叉污染。

I received a kit containing the MinElute columns; however, they were left out for a while and not stored at 2–8°C upon receipt. Can I still use them?

Short-term storage (up to 4 weeks) at room temperature (15–25°C) does not affect the performance. However, for optimal performance and quality, storage temperature should not exceed 25°C.

What can be used as an alternative to the A260 measurement for quantification of small amounts of RNA and DNA?

Small amounts of RNA and DNA may be difficult to measure spectrophotometrically. Fluorometric measurements, or quantitative RT-PCR and PCR are more sensitive and accurate methods to quantify low amounts of RNA or DNA.

Fluorometric measurements are carried out using nucleic acid binding dyes, such as RiboGreen® RNA Quantitation Reagent for RNA, and PicoGreen® DNA Quantitation Reagent for DNA (Molecular Probes, Inc.).
