therascreen KRAS Pyro Kit是用于鉴定KRAS基因中突变和缺失的分子检测试剂盒。该试剂盒包含扩增KRAS基因所需的引物和试剂，以及在PyroMark Q24实时定量焦磷酸序列分析仪上使用焦磷酸测序技术实时定量检测突变所需的缓冲液、引物和试剂。

将载有突变序列的质粒和野生型基因的质粒以四种不同突变质粒的比例：0、12.5、25、37.5和50%混合。每种质粒混合物四个重复样本随机放在板上分析。得出编码子12 GGT->TGT的突变结果。
精密度
编码子12 GGT->TGT的突变线性由3个不同的操作者在不同的三天使用不同的PyroMark Q24实时定量焦磷酸序列分析仪和试剂进行实验来确定。
检测0–50%突变范围，中间精密度（SD）的值为0.6–2.0%。

Principle
therascreen KRAS Pyro Kit用于实时定量检测人KRAS基因编码子12、13和61的突变，在PyroMark Q24实时定量焦磷酸序列分析仪上使用焦磷酸测序技术检测。KRAS基因编码在EGFR信号通路中有重要作用的蛋白。KRAS基因的突变会影响蛋白对下游信号通路的激活。在所有癌症中有将近30%发生KRAS基因突变，其中直肠癌（35%）和肺癌（18%）。
可检测以下种类的突变：
- 编码子12/13：G12A、G12C、G12D、G12R、G12S、G12V和G13D
- 编码子61：Q61E、Q61H、Q61L和Q61R
Procedure
该试剂盒包含2部分实验：一个用于检测编码子12和13的突变；另一个用于检测编码子61的突变（参见"Illustration of the KRAS assay"和"Pyrogram trace of a normal genotype in codons 12 and 13"）。运用PCR技术对这两个片段分别进行扩增，测定特定区域的核酸序列。使用该法进行定量和定性分析时，特定区域周围的核酸序列峰被视为标准峰和参照峰。
