绩效

使用FlexiGene体系纯化的DNA可达150 kb(参见" High-quality genomic DNA")。可用于扩增反应和Southern印迹实验,也可存档(参见" Efficient downstream PCR analysis")。

DNA在单管中通过离心操作,从0.1 – 20 ml(血液)、0.1 – 2 ml(白膜层)或1 – 2 x 106个细胞(培养细胞)中纯化DNA。1小时内处理12个样本。依据样本大小,再水合体积0.2 – 1 ml。

原理

FlexiGene DNA体系可从不同量的全血、白膜层和培养细胞中纯化基因组DNA。纯化过程在单管中进行,费用少,操作简单。纯化的DNA在多种下游应用中表现良好。

程序

首先在样本中加入裂解液(参见" FlexiGene DNA procedure")。细胞核和线粒体离心沉于底部,使用包含QIAGEN Protease的变性缓冲液重新悬浮。然后蛋白裂解,加入异丙醇沉淀DNA,离心处理,70%乙醇洗涤和干燥。使用水合缓冲液重新悬浮的DNA可即用于下游分析或–20°C贮存。可从全血、白膜层和培养细胞中获得高产量的DNA。

Do you have stability data for genomic DNA isolated with the Flexigene DNA Kit?

Yes. We have shown that DNA purified with the Flexigene DNA Kit is stable for at least 7 years when being stored at either 2-8°C or -20°C.  However, the DNA stability is dependent upon storage conditions.  The data is available here under Additional Resources.

Do you have a protocol for the isolation of genomic DNA from frozen clotted blood?

Yes, we have the following protocols:

  • Isolation of genomic DNA from frozen clotted whole blood using the QIAGEN Genomic-tip 100/G (QG02). TEST

You will need to prepare the required buffers according to the recipes in Appendix A of the QIAGEN Genomic DNA Handbook, or you can purchase the Genomic DNA Buffer Set containing pre-made solutions. Alternatively, the QIAGEN Blood & Cell Culture Midi Kit, containing Genomic-tips 100/G and buffers, can be used.

  • Purification of archive-quality DNA from clotted whole blood using Clotspin Baskets and the Gentra Puregene Blood Kit (PG03).
