Product Details

Investigator IDplex Plus Kit is a multiplex STR assay for human identification covering the 13 CODIS (Combined DNA Index System) core loci, D2S1338, D19S433 and Amelogenin.

The kits utilize our fast-cycling technology, enabling amplification in ca. 90 minutes. Highly sensitive with rapid and reliable results from trace DNA, the Investigator IDplex Plus Kit is suitable for all forensic applications and paternity testing. The genetic analyzer is calibrated with BT5 Matrix.

Investigator IDplex Kit meets ISO 18385 requirements.

Performance

Investigator IDplex Kit utilize fast-cycling technology to provide reliable results in STR assays in ca. 90 minutes from extracted DNA.

The Fast Reaction Mix is based on proven QIAGEN multiplex PCR technology that has been optimized to increase inhibitor tolerance. Assay performance tested in the presence of increasing concentrations of 6 types of inhibitor (humic acid, hematin, tannic acid, indigo carmine, calcium and collagen) shows high sensitivity and superior inhibitor resistance. This enables reliable, high-quality results from casework samples.

The Investigator IDplex Kit is validated and evaluated using the GeneAmp 9700 thermal cycler and the Applied Biosystems 3500 Genetic Analyzer.

The recommended amount of DNA under standard conditions for use with the Investigator IDplex Plus Kit is 0.5 ng. Internal validations demonstrated robust and balanced results with 0.2–2 ng DNA, and reliable results with <0.1 ng DNA.

Principle

Investigator IDplex Kit uses QIAGEN's fast-cycling PCR technology for the rapid, simultaneous amplification of the 13 CODIS (Combined DNA Index System) core loci, D2S1338, D19S433 and Amelogenin. Carefully designed primers reduce overlap of loci alleles, thereby minimizing the likelihood of misinterpretation.

Procedure

The 15 polymorphic autosomal STR markers (D2S1338, D3S1358, D5S818, D7S820, D8S1179, D13S317, D16S539, D18S51, D19S433, D21S11, CSF1PO, FGA, TH01, TPOX and vWA), and the gender-specific Amelogenin are amplified simultaneously in a single PCR and labeled with one of the dyes listed in the table "Investigator IDplex Plus Kit dyes and corresponding markers".

The amplified loci then undergo capillary electrophoresis. The results can be analyzed using dedicated software, such as GeneMapper ID-X. See the table "Technical specifications" for more information.

The BT5 matrix standard is designed for use with our Investigator STR and Dipplex Assays. We do not disclose the containing dyes.
