GeneRead Size Selection Kitは迅速で簡便なスピンカラム操作を用いた正確なサイズ選択およびDNAフラグメントの精製を実現します。このキットにはMinElute columnと、150 bp より長いDNAフラグメントの選択に至適化されたバッファーが含まれています。

原理

GeneRead Size Selection Kitは簡便なスピンカラムテクノロジーにユニークなデザインの結合バッファーを組み合わせています。MinElute spin columnは、あとから行なう反応のために高濃度の精製DNAを確実に提供します。キットに含まれる特別なバッファーは、アダプターやアダプターダイマーのような短いDNA断片を効果的に除去するように至適化されています。長いDNAフラグメントは適切な塩濃度によりシリカメンブレンに吸着し、短いフラグメントや夾雑物はカラムを通過します。これにより150 bpのカットオフ（電気泳動での視覚化により最短のフラグメント長が定義された）が行なわれます。直前の反応で用いた酵素などの不純物は効果的に洗浄され、純度の高いサイズ選択されたDNAが溶出されます。さらに、次世代シークエンスのためのライブラリー調製中のサイズ選択はアダプターモノマーおよびダイマーの的確な除去と、それに続く次世代シークエンス用のライブラリー調製の二つのステップで行なわれます。

操作手順

GeneRead Size Selection Kitは簡便なスピンカラム法を採用し、DNA ライブラリーの正確なサイズ選択を確保する一方、アダプターダイマーやモノマーの除去を効率的に行ないます。 QIAcube での自動化が可能で、使いやすいプロトコールは面倒なマニュアルでの作業を減らすことで時間を節約し、その後のアプリケーションの阻害物質となりうるエタノールあるいはビーズの混入のリスクがありません（図 ""）。

Short-term storage (up to 4 weeks) at room temperature (15–25°C) does not affect the performance. However, for optimal performance and quality, storage temperature should not exceed 25°C.

Full recovery is seen for fragments larger than 300bp. Between 150 and 300bp, recovery increases with size.

It is normal for the buffer to turn yellow and it does not impact the performance of the buffer.
